چکیده
Recently, a Na + -binding aptamer was reported to be embedded in few RNA-cleaving DNAzymes, including NaA43, Ce13d, and NaH1. The consists of multiple GG stretches, which is prerequisite for the formation G-quadruplex (G4) structures. These DNAzymes require activity but show no presence K or other metal ions. Given that DNA can selectively bind by forming G4 structure, this work aims answer whether also uses . Through comparative ThT fluorescence spectrometry studies, while control exhibited notable enhancement up 5 mM with d 0.28 ± 0.06 mM, Ce13d DNAzyme negligibly perturbed similar concentrations Opposed this, displayed specific remarkable decrease low millimolar NMR experiments at two different pH values suggest adopts significantly conformation equilibrium conformations versus has more stable structure Additionally, absence characteristic peaks one-dimensional 1 H not responsible binding. This hypothesis confirmed CD spectra sequence. Therefore, we concluded must selective it binds using structural element does contain G4.

Ethyl-substitutive Thioflavin T as a highly-specific fluorescence probe for detecting G-quadruplex structure
G-quadruplex has attracted considerable attention due to their prevalent distribution in functional genomic regions and transcripts, which can importantly influence biological processes such as regulation of telomere maintenance, gene transcription and gene translation. Artificial receptor study has been developed for accurate identification of G-quadruplex from DNA species, since it is importa...
متن کاملA Sensitive and Label-Free Pb(II) Fluorescence Sensor Based on a DNAzyme Controlled G-Quadruplex/Thioflavin T Conformation
Pb(II) can cause serious damaging effects to human health, and thus, the study of Pb2+ detection methods to sensitively and selectively monitor Pb(II) pollution has significant importance. In this work, we have developed a label-free fluorescence sensing strategy based on a Pb(II) DNAzyme cleavage and the ThT/G-quadruplex complex. In the presence of Pb(II), a G-rich tail was cut and released fr...

A Grafting Strategy for the Design of Improved G-Quadruplex Aptamers and High-Activity DNAzymes
Nucleic acid aptamers are generally obtained by in vitro selection. Some have G-rich consensus sequences with ability to fold into the four-stranded structures known as G-quadruplexes. A few G-quadruplex aptamers have proven to bind hemin to form a new class of DNAzyme with the peroxidase-like activity, which can be significantly promoted by appending an appropriate base-pairing duplex onto the...
متن کاملUltrafast fluorescence spectroscopy reveals a dominant weakly-emissive population of fibril bound thioflavin-T.
In this communication, using sub-picosecond resolved fluorescence upconversion spectroscopy, we discover that despite a large fluorescence enhancement observed for thioflavin-T in insulin fibrils, the majority of fibril bound thioflavin-T undergoes efficient ultrafast conformational relaxation, and thus does not contribute to the characteristic fluorescence enhancement.
